Internal delamination or burring in the line is causing an obstruction that seals tightly , but flattens against the wall under the greater weight of the larger tank load(or when vac is applied).
Have you dug up the line and checked it by section?

Another fun experiment, can you get 17 gallons to flow? If you get a partial drain, the water remaining in the tank will give you a good idea how far down the line any potential obstruction is.

If I run water up the tubing- instead of filling the tank from the top - it only takes a few gallons to get flow - as the entire length of tubing is full to start with.
It's only when the line is dry when starting. As far as digging up the line - not practical. The line runs inside a PCV tubing - and it's held in the center of that PVC by neoprene insulation. The whole system - tank and tubing - is insulated. Unrelated- Maple sap spoils when it gets above 45 degrees. We pick up the sap early in the morning - frequently with ice in the collection barrels. There is a thermometer in the tank - and I frequently see temps down to 29f - without freezing - due to sugar content.
Well, on the plus side, if it is delamination the hinged end will eventually fail and flake out through the line.
