Thumbs up button is for kudos; they attach to the post, and are meant to express that you liked the post for some reason
rep points (reputation points) can be given a user by clicking on the "rate" button on the right hand side of a post, or by going to the user's profile and clicking on the appropriate button under reputation (there you can also read the details of the reputation points the user has received, as well, which includes a one line comment)
You can go to a user's profile by clicking on their name in the upper left of a post. (their reputation report is also accessible via the green number representing the user's rep point total in the top right of a post)
