Point one: A happiest. An angriest. These are grammatical errors, and the repeated making of them becomes humorous to the CHINESE. Wouldn't want to give the Chinks a laugh, would you?
Point two: est is a suffix which indicates the highest degree/quality... superlative, observation of having most excellence.
Point three: the 'assiest' qualities would probably be found in an ass. Hence, either this is a really amusing suggestion that someone has a donkey in their head... or their head is an ass because their whole being is that of an ass and they are literally an ass. Regardless, your definition seems to occur in some alternate universe where an ass in a head is the clearest indication of assiness.
Point four: there is a point four, but I am feeling kind and will leave this one out.
