Once a player is drafted, no one else can have him (though most fantasy leagues allow teams to trade players). That's what makes the draft fun since you have to prioritize based on your position needs and which players are still available.
So no -- you and Luckie can't both have Manning.

For any fantasy sport, a group of people create "teams" that are stocked via a draft of some kind (which prevents players from being selected by more than one team). These teams are paired off against each other in the same way that pro sports leagues schedule the real games. The statistical value of the performances of the individual players are plugged into one or more formulas to produce a point total that is compared to that of the other team, so that at the end of the day a winner can be declared.
Some leagues use an auction-style draft where the participants are allotted a specific dollar amount to 'spend' on players. Deciding how much to "pay" a player, or how a trade might affect the team's "salary cap" and so on, can be more fun for them than the actual wins and losses.
